El sábado 21 de marzo de 2009 se llevó a cabo la sesión final de presentación de afiche por parte de los estudiantes que componen en programa Biominds de AMGEN. En el mismo, tuvimos la oportunidad de presentar los resultados obtenidos en cada una de las investigaciones de las cuales cada uno de los estudiantes ha participado. Fue una experiencia grata y sumamente enriquecedora ya que no solo pudimos demostrar nuestros conocimientos en nuestra área de estudio e investigación sino también de compartir con otros compañeros participantes del mismo programa que a lo mejor nunca habíamos visto o nunca habíamos tenido la oportunidad de hablar e intercambiar ideas. Esto se debió principalmente a que los estudiantes tuvimos la oportunidad de evaluar al menos tres afiches de distintos estudiantes participantes de programa.

De forma breve resumiré la evaluación de los mismos:

Afiche 1: Titulo: Isolation of important clinic yeasts in water treatment plants and Puerto Rico forest, de Isabelita Martínez. Su investigación es una muy interesante, ya que muestra cuáles levaduras clínicas estamos más expuestos los humanos, con la importancia de que estas al igual que las bacterias causan enfermedades mortales a las personas. La joven logró enfatizar la importancia de su investigación, logrando retener la atención de los demás. Presentó un buen afiche, de forma organizada y con concordancia.

Afiche 2: Titulo: Isolation and characterization of bioprospects from the Phytotelmata of the Pitcher Plant Nepenthes sp., de Josué G. Millán. Este afiche contenía información de manera organizada y completada en el sentido de poder transmitir el tema investigative de forma correcta. El joven tenía conocimiento extenso de su proyecto y por lo tanto logró demostrar además seguridad al presentar. Entiendo que es tuvo un excelente afiche y realizó una buena presentación, dado que además no tuvo problema alguno al contestar preguntas.

Afiche 3: Titulo: In vitro Culture of Foeniculum Species, a Traditional Medicinal Plant, de Verónica Figueroa.  Este trabajo fue presentado de forma organizada y coherente.  En cuanto a su investigación, a pesar de habérsele presentado problemas con la planta, logró obtener resultados a presentar.  Entiendo que le faltó un poco de seguridad al hablar, lo que podría ser por los nervios, pero hizo un buen trabajo.  Me pareció interesante además, el hecho de que ese trabajo nació por iniciativa de la estudiante, debido a su curiosidad por las propiedades medicinales de la planta.

Isolation and characterization of organic compounds from Momordica charantia “Cundeamor” Infusion used for Diabetes Type II in Puerto Rico

Momordica charantia L., locally known as Cundeamor, is being widely used as tea to decrease blood glucose levels of diabetic patients, according to popular claims. After doing a scientific literature review, evidence has been found of numerous works validating the hypoglycemic activity of variations of the same species in other countries. This can imply the presence of similar chemical composition among variations of the plant. Currently, our work presents the first stage of the long term goal, which is the process of isolation and characterization of potentially bioactive compounds from the infusion of Cundeamor and eventually comparative studies of the chemical composition previously reported for variations of the same species. As part of the methodological strategy, the homemade preparation of Cundeamor, the infusion, and the methanolic Sohxlet extract of the leaves were studied.  The aqueous solutions of the infusion were extracted with solvents of different polarities, and the components of the crude extract were isolated by column chromatography. Identification of organic components in every extract was performed by means of thin layer chromatography, TLC. Optimization of the developing solvents mixtures for improving identification of the components in each extract and the reproducibility of the analysis will be reported. Furthermore, preparative TLC results of different fractions will be reported.  Spectroscopic analysis will be presented for those compounds isolated successfully.

De los objetivos establecidos en enero podría decir que prácticamente he alcanzado un 50% del progreso. Debido ha que he estado optimizando la metodología al momento no me he encontrado con dificultades experimentales, excepto en el sentido de que los materiales han comenzado a limitarse según la investigación lleva más de un año en progreso. Mientras, espero lograr los objetivos finales.

Este es el ultimo semestre en el que estare trabajando para BioMinds. Mi mentora y yo esperamos lograr obtener analisis de identificacion y caracterizacion mas definidos.

Para ello trabajare con las infusiones de la planta Cundeamor y realizare comparacion con extarccion sohxlet. Esto nos permitrira ademas, realizar comparacion con la literatura.

Se continuara optimizando la metodologia basandonos en los resultados del semestre pasado, ya haciendo enfoque en extractos especificos de diclorometano y eter.  Se trabajara con distintos metodos de caracterizacion como FT-IR y RMN. Tambien con TLC preparativo.

Esperamos adelantar trabajo para lograr presentar esta investigacion en la conferencia nacional de American Chemical Society (ACS) en Utah.

During this semester I was able to work with three different extracts of Cundeamor: the crude extract, the Sohxlet extract and the infusion. My results show that organic compounds are being extracted in each by solvents of different polarities. Also, as expected, a major quantity of constituens are extracted in the crudes. We should work on the quantity of solvents used since we found that the quantity extracted are being limited by the solvent’s. With the spectroscopic results of NMR we found that the Sohxlet extract compares with the infusion by identifying similar picks in the extracts. More trials will be done to optimize the quantities extracted by the quantity of leaves and solvent used. As future work, we are interested in performing preparative TLC to purify the extracts and do more spectroscopic analysis. Also, we should study the bioactivity of the extracts by performing bioassays on antidiabetic effects.

In the scale of 1 to 5, this time I describe my project with a 4 by having more or less 80% of our objectives accomplished. I have optimized most of the mixture of solvents and we have analyzed most of the sample extracts by NMR. Also, I’ve been optimizing the quantities of leaves used for this project. Furthermore, we will optimize the isolation of components by column chromatography and we will need some more spectroscopic analysis of samples. Finally, we hope to perform the preparative TLC technique.

With our findings we can identify the type of components that are present in the infusion of Cundeamor that could be responsable for the claimed effects on type II diabetes.

During this semester I had progress in my project and hence I have learned a new technique. This technique is column chromatography and we use it to isolate compounds from the plant Cundeamor extract, to later analyze by spectroscopic methods. In column chromatography, the stationary phase, a solid adsorbent, for which we use silica, is placed in the column and the mobile phase, a liquid, for which we use various solvents adding by polarity, is added to the top and flows down through the column by gravity.

In the scale 1 to 5, I believe I have a progress of 3 which describes a 50% of the objectives described previously. I’ve been working out the mixture of solvents for optimal separation and has I mentioned before I also have been working with isolation and characterization methods. I still need to learn the technique of preparative TLC and apply all of these to our three principal extracts of Cundeamor: the infusion, the leaves crushed in methanol and the crude by Sohxlet.

Lately, I have encountered some difficulties for the progress of my project because of time issues, because of some random days off that were not part of the academic calendar, and there is not much to do in those cases but to make sure of great performence during the working days.

During the first semester I was able to work out the infusion and crude extract of the plant under study. I basically developed the first stage of the methodology but I still need to optimize the method for identification of compounds in each fraction by selecting the best mixture of solvents by TLC. This is an important step since our goal for this semester is to characterize the compounds of interest by NMR. After having the optimal mixture of solvents I will be working with the separation and purification of compounds by Preparative TLC and Column Chromatography. We are also searching for an appropriate bioassay that will be performed for each fraction to identify if there is any bioactivity present.

This past summer I went to the North Carolina State University to do some research in the Chemistry Department at Alexander Deiter’s Lab. My project consisted of developing a methodology to rapidly synthesize non-natural amino acids by using microwave irradiation. It was a wonderful experience, were I was able to apply some of my Organic Chemistry background and also, learn a lot of new concepts and techniques. I was a participant of the AGEP-SRE Program which is sponsored by the NSF.

Durante este semestre he tenido la oportunidad de aprender varias técnicas de laboratorio, las cuales ya he mencionado. Pero creo que también es muy importante el hecho de adquirir experiencia en el laboratorio de forma individual, ya que de esta forma uno aprende más a tener pensamiento crítico y analizar de forma independiente; claro está, con la ayuda del mentor.

En cuanto a barreras, entiendo que no me he encontrado con nada dificultoso. Tal vez, haber tenido experiencia previa en investigación me ha ayudado mucho. Pero sí entiendo que tomando en cuenta que estoy en medio del desarrollo inicial de mi proyecto de investigación, no he tenido el tiempo suficiente para obtener cómodamente mis resultados, ya que dentro de pocos días nos toca presentar y aún queda mucho trabajo por hacer. Pero ya estoy trabajando en esto y espero que en el final del camino todo quede como he esperado desde principios del semestre.

Definitivamente que para el próximo semestre sé que el trabajo será más llevadero, ya que comenzaré con una metodología que he estado desarrollando. Pero además, tendré la oportunidad de aprender técnicas nuevas según avance mi proyecto. Por tanto, entiendo que la parte más difícil siempre será el comienzo, pero eso lo confirmaré el próximo semestre.
